Experimental Process

The promoter of lrp was mapped with primer extension. Regulation by Lrp was shown by plrp-lacZ reporter assays comparing w-t and lrp mutant, using truncations and site-directed mutagenesis of the promoter region containing putative sites. Binding was shown first with EMSA on truncated plrp fragments and then confirmed with DNAse footprinting.
