Experimental Process

A CsoR homologue in S. aureus was identified by computational search (36% identical to the B. subtilis CsoR). Total RNA was extracted from wild-type and CsoR mutants and analyzed by Northen blot to show that CsoR is responsible for copper-dependent repression of copAZ and copBmco operons, and their operon configuration confirmed. The expression of other genes under low and high copper conditions in the w-t and CsoR mutant was investigated by RT-PCR, revealing repression of copAZ and clpC.
