Experimental Process

Proteome analysis identified a 3-fold derepression of ycgT expression in fur mutant cells compared to the wild-type cells. Inspection of the ycgT promoter region identified a putative Fur-binding site. Binding of Fur to the target promoter was shown by EMSAs. ycgT::lacZ transcriptional fusion confirmed that ycgT expression is repressed by Fur.
