Experimental Process

Sites were identified by fusing different length promoter fragments with a promoterless LacZ and comparing B-gal activity. The results of these assays suggest the presence of 3 sites in the ompU promoter. The existance of these sites was verified with emsas of the promoter fragments, and a DNase protection assay, which showed three footprints. The authors do not attempt to narrow down the sites further.
