Experimental Process

Regulatory impact of IFH was determined via B-gal assay. A DNase I assay showed that IHF bound a region in the tcpA promoter, overlapping the footprint of H-NS (relieving H-NS repression is a known method of action for IHF). Two IHF binding sites at -162 and -35 (based on the E. Coli IHF consensus site) were found in the protected region; EMSA analysis of promoter fragments indicated that IHF bound the -162 site independently of the -35 site, but could not bind the -35 site alone.

The paper reports that TF forms complex with other proteins for binding with reported sites.

IHF is a heterodimer, consisting of ihfA (YP_001216792.1), and ihfB(YP_001217447.1)
