Experimental Process

Regulation for both promoters demonstrated by beta-gal assay comparing to a codY-null mutant in aa rich medium. CodY sites were identified by visual comparison to known consensus on DNAse footprinted regions. The functionality of sites was further verified by site-directed mutagenesis, followed by beta-gal assays and DNAse footprinting.
