LacZ reporter assays in wild-type and mexT mutant strains showed that MexT is necessary for expression of the mexEF-oprN operon. Promoter deletion analyses with lacZ reporter assays were done to identify the promoter region that contains MexT binding sequence. Promoter deletion analysis with EMSA was done to localize the MexT binding site. Site-directed mutagenesis of the putative MexT binding site in conjunction with lacZ reporter assay and EMSA confirmed the specific interaction of MexT with its target site.
