Experimental Process

A consensus search identified the putative binding site. Site expression was induced in PhoB variable strains via phosphate starvation, and GUS assay indicated expression dependence on PhoB. A Western blot of PhoB variable cell extracts confirmed a loss of expression in the absence of PhoB. EMSA identified strong binding to the putative site.
