Experimental Process

sypA, sypI, sypM, sypP - lacZ reporter assays confirmed that SypG has an important role in the expression of these genes. lacZ reporter assays with wild type and truncated promoters of the syp genes showed that promoter truncation downstream of the previously reported putative SypG sequence resulted in a severely diminished syp transcription. The effect of putative SypG binding site on the expression of sypA was evaluated by monitoring the development of the spot from a smooth to wrinkled morphology over time. A biofilm formation assays with site-directed mutagenesis confirmed that SypG binding sequences in the promoters of syp genes was important for their expression. ChIP analysis in conjunction with qRT-PCR showed that SypG binds to syp promoter regions to activate syp transcription. Regulatory Sequence Analysis Tools (RSAT) program identified three additional SypG targets that contained both a putative SypG binding site and a putative downstream σ54 binding site.
