Experimental Process

A cDNA microarray of S. coelicolor ORFs identified over 300 S. lividans genes affected by adpA, either activated or repressed. Of these, six genes relating to development, the cell envelope, primary or secondary metabolism, or secreted proteins were examined in more detail by qRT-PCR, all of which showed down-regulation in the adpA mutant. The S. griseus consensus binding site was used for a PSSM analysis which identified between three and nine putative binding sites for each of the six genes. EMSA was used to confirm or refute each putative site, and it was found the six genes had between three and five binding sites in their respective promoter regions.
