Experimental Process

The authors identified a 20-bp ExpR consensus sequence from an alignment of seven previously confirmed ExpR binding sites. This consensus was then used to search for addition ExpR targets in the S. meliloti 1021 genome. EMSAs with DNA probes containing the predicted ExpR binding sites confirmed the ExpR targets sites identified by the authors. Site-directed mutagenesis and EMSA showed that the modification of the several predicted binding sites (C to T) resulted in the abolishment of binding by ExpR. Promoter-gfp reporter assays with selected promoters bound by ExpR revealed ExpR-mediated expression regulation.
