Experimental Process

A putative ArgR binding site in the arc promoter was identified by visual inspection. This binding site was similar to the previously identified ArgR binding sites. Site-directed mutagenesis of the predicted ArgR binding site resulted in the loss of arginine induction as determined by ADI measurements. EMSA confirmed that ArgR binds to the arcD promoter. This binding was abolished when the binding site was mutated. DNase I footprinting localized the ArgR binding region. Sequence alignment of ArgR binding sites was used to identify the ArgR consensus, 5'-TGTCGCN8AA-3'.
